摘要:For long-term sustainability of wild service tree genetic resources, micropropagation technologies could prove to be useful (Dujíčková et al. 1992, Chalupa 1992, Malá et al. 2005). For standardized wild service tree procedures of micropropagation and for its effective exploitation in the forestry, some difficulties during rooting stage of organogenesis remain to be solved. In this study we compared the influences of three different aromatic cytokinins BAP, mT, and MeoBAPr on the multiplication of explants and the rooting of microcuttings of the wild service tree. The highest multiplication rate shows using of multiplication medium supplemented with BAP. Only 55 - 60% of multiplication rate with BAP was observed in media containing MeoBAPr and mT. Numbers of successfully rooted plantlets were determined after 6 weeks of cultivation in the rooting medium. The highest numbers of rooted plantlets were achieved after transferring of the microcuttings from medium with MeoBAPr into one-third MS medium with NAA. We studied the influence of cold treatment during multiplication, 85% of rooting rate of microcuttings of the wild service tree in this condition were reached.
