摘要:The European aspen (Populus tremula) represents a very modest tree species in climatic conditions and soil quality. For its regeneration ability, it is a very perspective woody species for tissue culture manipulation, genetic engineering, and genetic mapping. Stem segments and leaves discs of the hybrid aspen (Populus tremula x P. tremuloides) clone no. 5 preserved in the FGMRI gene bank were used for testing of genetic transformation by agrobacterium tumefaciens. Transformed shoots were obtained using four agrobacterial constructs bearing uidA (GUS) gene governed by various promoters. Presence of nptII and GUs genes in transgenic tissues and plants was verified using the polymerase chain reaction.
