摘要:At present,there is not much research on pro-teomics in the field of postharvest research on fruits and vegetables.In some research groups,post-harvest induction of resistance in fruit trees is a promising technique.In order to obtain a clear protein gel map,this paper establishes an optimization system for two-dimensional electrophoresis of pear leaf protein.Optimize the steps of protein extraction method,hydration operation mode of the strip,setting of isoelectric focusing (IEF) program,and protein loading.The results showed that the homogenate method achieved good results in pear leaves in different environments.There were fewer vertical and horizontal striations and more protein spots can be observed by naked eyes in the whole electrophoresis map,and the distribution of points is even and clear.The experimental method in this paper effectively improves the resolution and lays a foundation for the differential proteomics research of pear leaves in different environments.
