  • 摘要:Miniaturization of explants allows reduction in space requirements and consequently labour cost for the maintenance of germplasm collections. Protocol was developed for the slow growth of date palm in vitro tissue for medium term storage and the viability of such tissues were analysed. Genetic fidelity of plantlets regenerated from short and medium term stored date palm tissues was analysed using appropriate molecular markers. The highest survival rate (80%) and switch to plants (75%) were observed with 1.2 M sucrose. To determine the possibility of cryoprotectant solution on freezing tolerances, cultures were exposed to dissolve the cryoprotectant solution for 20-100 minutes. The maximum survival rate was obtained with exposed cultures. Random Amplified Polymorphic DNA (RAPD) was extensively used to study the genetic stability of cryogenic tissue cultures from date palm. According to RAPD analysis, the seedlings derived from cryopreserved cultures were similar to those derived from untreated cultures, both of which were similar to the cultivated plants in the field. Finally, all these aspects were contributing the improvement of currently available techniques for in vitro germplasm conservation of date palm and whole plants of chilled cultures were able to adapt to free living conditions after a stage of adaptation. The aim of the study was the collection and in vitro germplasm preservation of the Alahsa date palm cultivars and was developed a date palm tissue culture media with minimal level of somatic variation.
