摘要：Alakline phospahatase belongs to the class of hydrolases and catalyzes the hydrolysis of a number of phosphoric acid esters, nucleotides etc. Extracellular Alkaline phosphatase (AlPase) was isolated from Pseudomonas sp., purified to near homogeneity with ion exchange chromatography on DEAE- cellulose followed by gel filtration chromatography on sepharose 6B. Characterization studies revealed that the enzyme molecular mass was 66 kDa. The purified enzyme exhibit pH optima at 10.0 and it was stable at pH range of 10-11, optimal temperature was determined to be 50°C with thermal stability over a range of temperatures from 40 to 60°C.
